This study aimed to investigate the major bioactive compounds, chemical constituents, and antioxidant activities of leaf extracts obtained from three cultivars of Tamarindus indica L. commonly cultivated in Phetchabun Province, Thailand, namely Sri Chomphu, Priao Fak Yao, and Si Thong, as well as to develop a prototype herbal shower cream containing tamarind leaf extract. The leaves were extracted using ethanol as the extraction solvent. The results revealed that the Sri Chomphu cultivar exhibited the highest total phenolic content 346.60 ± 0.40 mg gallic acid equivalent (GAE)/g extract), followed by Priao Fak Yao 334.66 ± 4.77 mg GAE/g extract and Si Thong 327.69 ± 0.39 mg GAE/g extract. Similarly, the highest total flavonoid content was observed in the Sri Chomphu cultivar 704.60 ± 0.58 mg quercetin equivalent (QE)/g extract, followed by Priao Fak Yao 626.02 ± 1.04 mg QE/g extract and Si Thong 559.82 ± 1.15 mg QE/g extract. High-performance liquid chromatography (HPLC) analysis identified gallic acid at concentrations of 88.23, 57.33, and 32.99 μg/g extract, respectively, while quercetin contents were 82.38, 39.07, and 32.80 ?g/g extract, respectively.
The antioxidant activities of the extracts were evaluated using the ferric reducing antioxidant power (FRAP) assay and the DPPH radical scavenging assay. The FRAP values were 203.98 ± 0.12, 166.21 ± 0.12, and 157.37 ± 0.12 mg Trolox equivalent (TE)/g extract for the Sri Chomphu, Priao Fak Yao, and Si Thong cultivars, respectively. Likewise, the DPPH assay demonstrated antioxidant capacities of 159.93 ± 0.22, 141.57 ± 0.99, and 130.65 ± 0.21 mg TE/g extract, respectively. Furthermore, the half-maximal inhibitory concentration (IC50) values were 0.31, 0.33, and 0.39 μg/mL, respectively. A lower IC50 value indicates a greater antioxidant capacity because a smaller amount of extract is required to scavenge 50% of DPPH free radicals. Accordingly, the Sri Chomphu cultivar exhibited the strongest antioxidant activity among the three cultivars. This finding was consistent with its higher levels of total phenolic compounds, total flavonoids, gallic acid, and quercetin, suggesting a close association between phytochemical composition and antioxidant potential.
Overall, the findings demonstrate that the Sri Chomphu cultivar is a promising natural source of antioxidant compounds and is an appropriate candidate for the development of a prototype herbal shower cream. This study provides scientific evidence supporting the utilization of tamarind leaf extract as a functional ingredient in herbal cosmetic formulations, contributing to the value-added utilization of local medicinal plant resources and facilitating their further development into commercially viable natural cosmetic products.
