This research aimed to prepare graphene oxide from waste silk floss generated from silk production and to investigate its potential application as an electrode-modifying material for electrochemical sensors. The study was conducted in two main stages: preparation and characterization of graphite and graphene oxide/reduced graphene oxide derived from waste silk floss, and application of the prepared material for the development of an electrochemical sensor for quercetin detection. The results showed that waste silk floss could be converted into insoluble silk fibroin powder and subsequently transformed into carbon-based materials, including graphite and graphene oxide/reduced graphene oxide. SEM analysis revealed morphological changes from fibrous structures to carbonaceous sheet-like materials with layered structures. SEM-EDS results indicated that the prepared graphite mainly consisted of carbon and oxygen, with atomic percentages of 92.72% carbon and 6.90% oxygen, while the reduced graphene oxide contained 96.63% carbon and 2.11% oxygen. FTIR spectra confirmed the presence of important functional groups, including O-H, C=O, C-O, C=C, and C-C, which are associated with carbon-based and graphene oxide structures. XRD analysis also revealed diffraction peaks corresponding to graphite and graphene-related structures, confirming that the preparation process successfully converted waste silk floss biomass into carbon materials suitable for electrochemical applications. Electrochemical characterization showed that the prepared graphene exhibited higher capacitance than graphite over almost the entire frequency range, indicating better charge storage capability and more pronounced capacitive behavior. The prepared graphene oxide was then applied to modify pencil graphite electrodes, resulting in three types of working electrodes: PGE, Chitosan/PGE, and GO-Chitosan/PGE. Cyclic voltammetry and electrochemical impedance spectroscopy performed in 5.0 mM [Fe(CN)6]3-/4- containing 0.1 M KCl demonstrated that the GO-Chitosan/PGE exhibited higher current response and lower charge-transfer resistance than the other electrodes. These results indicate that graphene oxide improved the electrochemical performance of the electrode by increasing the electroactive surface area, providing additional active sites, and enhancing electron transfer at the electrode surface. The electrodes were further evaluated for quercetin detection using differential pulse voltammetry. Among the tested electrodes, GO-Chitosan/PGE showed the best analytical response, with an Ipa corrected value of 0.445 mA and an S/B ratio of 2.41, which were higher than those of PGE and Chitosan/PGE. Quantitative analysis of quercetin over the concentration range of 0–50 μM provided a calibration equation of (y = 2.4144x + 378.31), with an (R2) value of 0.9845 and a sensitivity of 2.4144 μA/μM. The limit of detection and limit of quantification were 8.727 μM and 29.091 μM, respectively. In addition, the GO-Chitosan/PGE exhibited good repeatability, with a %RSD of 1.51, acceptable reproducibility, with a %RSD of 7.04, and acceptable stability after 7 days of storage, retaining 81.30% of its initial signal. In conclusion, this study demonstrates that waste silk floss can be used as a starting material for the preparation of graphene oxide/reduced graphene oxide. The prepared material showed potential as an electrode-modifying material for electrochemical sensor applications. In particular, the GO-Chitosan/PGE exhibited promising performance for quercetin detection at the micromolar level, together with acceptable repeatability, reproducibility, and stability. These findings support the feasibility of valorizing silk waste into functional carbon-based materials for further development of electrochemical sensor platforms.
