การพัฒนา และตรวจสอบความใช้ได้ของวิธีวิเคราะห์หาปริมาณสารสาคัญในสมุนไพร ฟ้าทะลายโจร (สารแอนโดรกราโฟไลด์ และดิออกซีแอนโดรกราโฟไลด์) ในตัวอย่างสมุนไพร ฟ้าทะลายโจรและผลิตภัณฑ์จากสมุนไพรฟ้าทะลายโจร ที่มาจากจังหวัดเพชรบูรณ์ 10 ตัวอย่าง วิเคราะห์โดยเครื่องโครมาโทกราฟีของเหลวสมถรรนะสูง (HPLC) ในการแยกใช้คอลัมน์ ZORBAX Eclipse Plus C18 (4.6×100 mm, 3.5 μm) ที่อุณหภูมิ 30 องศาเซลเซียส ด้วยเฟสเคลื่อนที่อัตราส่วนไม่คงที่คือน้ากับอะซิโตไนไตรล์ (70:30, v/v) ที่อัตราการไหล 1 มิลลิลิตรต่อนาที ใช้ตัวตรวจวัดแบบ Diode Array Detector (DAD) ที่ความยาวคลื่นของ Andrographolide 223 และ 14-DDA 252 นาโนเมตร ตามลาดับ ในการทดสอบความจาเพาะเจาะจงนั้น พบว่าพีคของสารทั้ง 2 ชนิดนี้สามารถแยกออกจากสิ่งรบกวนได้ดี ช่วงความเป็นเส้นตรงทั้ง 2 ชนิดอยู่ในช่วง 10.00 - 250.00 และ 1.00 - 30.00 ไมโครกรัมต่อมิลลิลิตร ค่าสัมประสิทธิสหสัมพันธ์เท่ากับ 0.9993 และ 0.9999 ตามลาดับ ในการทดสอบความแม่นได้ร้อยละการกลับคืน (%recovery) อยู่ในช่วง 90-100 ค่าร้อยละเบี่ยงเบนมาตรฐานสัมพัทธ์ (%RSD) ไม่เกิน 2 วิธีวิเคราะห์ที่พัฒนาขึ้นมานี้มีความคงทนต่อการเปลี่ยนแปลงจากสภาวะต่าง ๆ ซึ่งถือว่าประสบความสาเร็จ และเป็นวิธีที่รวดเร็ว, ง่าย, ราคาถูก, มีประสิทธิภาพ, ทนทาน และปลอดภัย
Development and validation of analytical methods for quantifying the essence in Andrographis paniculata (Andrographolide and deoxyandrographolides) samples in Andrographis paniculata and Andrographis paniculata herbal products. 10 samples from Phetchabun province were analyzed using high-performance liquid chromatography (HPLC). The ZORBAX Eclipse Plus C18 column (4.6×100 mm, 3.5 μm) was isolated at 30 °C. A mobile phase gradient elution was water to acetonitrile (70:30, v/v) at a flow rate of 1 ml/min. Use a diode array detector (DAD) at the wavelength of Andrographolide 223 and 14-DDA 252 nm, respectively. These two substances' peaks can be separated from the interference well. The linearity ranges for both species ranged from 10.00 - 250.00 and 1.00 - 30.00 μg/mL. The correlation coefficients were 0.9993 and 0.9999, respectively. The accuracy test percentage recovery ranged from 90-100. The relative standard deviation percentage (%RSD) was less than 2. This developed analytical method was tolerant to changes in various conditions, which was considered successful. Moreover, it's fast, easy, cheap, efficient, durable and safe.
